De Minaur's 6-4 3-6 6-3 hard-court win in the 250 series event maintained the Australian's 100 per cent record against Paul, stretching it to five wins in five meetings with the world No.14.
As the fifth seed continues to fine-tune his preparations for the US Open, which begins at Flushing Meadows on August 28, de Minaur will face Germany's Dominik Koepfer for a place in the final.
The No.7 seed dispensed with the USA's Aleksandar Kovacevic in straight sets, 6-4 7-5.
Greek top seed Stefanos Tsitsipas takes on Chile's Nicolas Jarry for a place in the other semi, against either Croatia's Borna Coric or Belarusian Ilya Ivashka.
